Chemical Stabilization and Characterization of PC645 Conjugates from Chroomonas placoidea

Abstract
Chemical stabilization and crosslinking of phycocyanin 645(PC645)from cryptophytes are achieved by using the small molecule crosslinking agent formaldehyde.It is found that the crosslinking reaction does not affect the fluorescence properties of the complex,but initially affects the 645 nm absorption peak associated with the PCBβ82 chromophore in the crosslinked product,followed by the 580 and 625 nm characteristic peaks.Based on the molecular weight indicators of phycoerythrin hexamer from Polysiphonia urceolata and PC645 heterodimer from Chroomonas placoidea,the molecular weight of the stabilized crosslinked product is preliminarily determined by a molecular sieve chromatography system to be about 120 ku,which is formed by two heterodimers.Characteristic studies show that the crosslinked product is not completely dissociated by SDS-PAGE,IEF,or at a low salt concen-tration of 0.05 mol/L PBS,and exhibits high tolerance to high concentrations of urea,SDS,and mercaptoethanol.The crosslinked product maintained basic spectral characteristics during a 3-month storage period,showing a half-life of more than 3 months.The experimental results lay a foundation for the development of a new type of chemical-ly stabilized phycobiliprotein fluorescent probe from cryptophytes in the future.关键词
