吡仑帕奈单药治疗伴中央颞区棘波的自限性癫痫的疗效和安全性
Comparison of the clinical efficacy of perampanel monotherapy in self-limiting epilepsy with spikes in the central temporal region

Abstract
AIM To investigate the clinical efficacy and safety of perampanel(PER)monotherapy in the treatment of self-limited epilepsy with centrotemporal spikes(SeLECTS).METHODS A total of 100 cases of children with SeLECTS in the hospital from May 2022 to January 2023 were randomly divided into the observation group(48 patients treated with oral PER)and control group(52 patients treated with oral levetiracetam).The clinical efficacy,adverse reactions and changes in the slow waves index(SWI)of nonrapid eye movement(NREM)sleep on electroencephalogram(EEG)were observed and compared after 6 and 12 months of treatment between the 2 groups.RESULTS Regarding seizure control,the effective rates of seizure control in the observation group were 54.27％and 77.08％after 6 months and 12 months of treatment,respectively.In the control group,the effective rates of seizure control were 40.38％and 51.92％respectively.Moreover,after 12 months of treatment,the effective rate of seizure control in the observation group was higher than that in the control group(P＜0.05).Regarding the SWI,there were no statistically significant differences in the improvement rates of SWI between the 2 groups after 6 months and 12 months of treatment.However,after 6 months of treatment,the improvement rate of SWI in children with spike-slow wave activation during slow-wave sleep in the observation group was higher than that in the control group(P＜0.05).Regarding adverse reactions,there were no statistically significant differences between the 2 groups after 6 months and 12 months of treatment.Regarding the retention rate of drug treatment,through follow-up,it was found that the retention rates of drug treatment in both groups were 100.00％after 12 months of treatment.CONCLUSION PER monotherapy is beneficial for early improvement of SWI in NREM sleep in pediatric patients with SeLECTS,which may enhance clinical efficacy.PER monotherapy for SeLECTS demonstrates a favorable efficacy and safety profile,making it a promising treatment option worthy of clinical promotion.关键词
