文成糯米山药染色体核型分析及基因组大小预估
Chromosome karyotype analysis and genome size estimation of Wencheng glutinous rice yam

Abstract
Flow cytometry was used to analyze the number and morphology of chromosomes in Wencheng glutinous rice yam,while flow cytometry was employed to identify the chromosomes karyotype and estimate its genome size.Using young stem segments of Wencheng glutinous rice yam as experimental materials,flame drying method was employed to obtain mitotic metaphase chromosome sections.DAPI staining fluorescence observation was used to count the chromo-somes,and then the karyotype was analyzed.Diploid shield leaf yam tender leaves and com(B73)were used as internal references,and flow cytometry was applied to determine the genome size and ploidy of Wencheng glutinous rice yam.The results showed that the number of chromosomes in Wencheng glutinous rice yam was 40,with a karyotype formula of 2n=2x=40=40 m,classified as lB type,representing a relatively primitive evolutionary type.The genome size of Wencheng glutinous rice yam was estimated to be 0.59 Gb.This results provide a basis for subsequent genetic improve-ment and breeding.关键词
