摘要

Abstract

Objective:To investigate the inhibitory effect and the mechanism of influence of Qiguiyin(QGY)Decoction on macrophage M1 polarization in cecal ligation and puncture(CLP)acute lung injury mice through JAK2/STAT3 signaling pathway.Methods:In this study,100 C57BL/6 mice were used and randomly divided into 20 mice each in the sham-operation group(sham group),model group,and QGY Decoction low,medium,and high dosage groups(QGY-L,QGY-M,and QGY-H groups).The sham group was operated only with open abdomen,and the rest of the groups were perforated by CLP to establish the septic lung injury model.The QGY-L,QGY-M,and QGY-H groups were gavaged with Chinese medicinal liquid at 6 h after the operation in the model group,and the sham group was gavaged with pure water at the same time point.Samples were collected 24 h after modeling to compare the pathological changes in lung tissues,Smith scoring for lung injury,and wet/dry weight ratios among the groups to determine the relatively optimal concentration of the QGY Decoction.The levels of iNOS and CD206 in bronchoalveolar lavage fluid were measured by flow cytometry.The protein expression levels of p-JAK2,JAK2,p-STAT3,and STAT3 in lung tissues were detected by Western blotting.The expressions of inflammatory cyto-kines IL-6,IL-1β,TNF-α,and IL-10 in mouse serum were measured using a high-throughput liquid phase pro-tein chip.Results:CLP successfully established a model of sepsis-induced lung injury.Compared with the sham operation group,lung tissue damage was more severe in the model group,with increased levels of iNOS and CD206(P<0.05),significantly upregulated p-JAK2/JAK2 and p-STAT3/STAT3 expressions(P<0.05),and markedly ele-vated serum inflammatory cytokines IL-6,IL-1β,and TNF-α(P<0.05),while there was no significant change in IL-10 expression.Compared with the model group,lung tissue damage was less severe in the QGY groups,with re-duced iNOS expression,increased CD206 expression,significantly downregulated p-JAK2/JAK2 and p-STAT3/STAT3 expressions,and markedly decreased serum inflammatory cytokines IL-6,IL-1β,and TNF-α,but signifi-cantly increased IL-10 expression(all P<0.05).Conclusion:QGY Decoction can alleviate CLP-induced sepsis acute lung injury by downregulating JAK2/STAT3 phosphorylation,thereby inhibiting M1 macrophage polarization.
