芩百清肺浓缩丸通过TNF-α/NLRP3/Caspase-1信号通路对感染后咳嗽大鼠气道高反应性的影响
Qinbai Qingfei Concentrated Pill's Impact on Airway Hyperresponsiveness in Post-infectious Cough Rats via the TNF-α/NLRP3/Caspase-1 Signaling Pathway

Abstract
Objective:To observe the impact of Qinbai Qingfei Concentrated Pills(Qinbai)on the TNF-α/NL-RP3/Caspase-1 signaling pathway,thereby alleviating airway hyperresponsiveness in post-infectious cough(PIC)rats.Methods:40 SD healthy male rats were divided into four groups:blank control group,model group,Qinbai group,and positive control group.The models were established using a combination of smoking,LPS solution nasal drops,and capsaicin solution nebulization inhalation.Cough frequency was observed;ELISA was used to measure the content of TNF-α in serum and bronchoalveolar lavage fluid(BALF);HE staining was performed to observe pathological changes in lung tissue;RT-qPCR and Western blotting were used to measure the expression of NL-RP3 and Caspase-1 in lung tissue.Results:Compared with the model group,the frequency of cough significantly decreased in the Qinbai group(P<0.01);the content of TNF-α in serum and BALF also significantly decreased(P<0.01);bronchial epithelial injury and inflammatory response were improved;the mRNA levels and protein ex-pression of NLRP3 in lung tissue were significantly reduced(P<0.01),the mRNA level of Caspase-1 was reduced and its protein expression was significantly reduced(P<0.05 or P<0.01).Conclusion:Qinbai Qingfei Concentrat-ed Pills can significantly reduce airway hyperresponsiveness in PIC rats,and its mechanism of action may be relat-ed to the TNF-α/NLRP3/Caspase-1 signaling pathway.关键词
