Identification,Biological Characteristics and Botanical Pesticide Screening of the Pathogen of Black Spot on Paeonia lactiflora

Abstract
Objective:To clarify the pathogen species and biological characteristics of black spot disease of Paeonia lactiflora in Xinyang area,and screen the suitable botanical pesticide.Methods:The Paeonia lactiflora diseased leaves were collected from the plant-ing area,the pathogenic bacteria were isolated and purified in the laboratory,and the pathogenic back inoculation test was carried out.The pathogenic strains were identified by morphology and molecular biology.By studying the biological characteristics of the pathogen,the optimal culture conditions were determined.The toxicity of three botanical pesticides to black spot of Paeonia lactiflora was deter-mined by mycelial growth rate method.Results:Combined with the analysis of morphology and molecular biology identification based on rDNA-ITS sequence and tef1 genes,the pathogen causing black spot disease of Paeonia lactiflora in Xinyang area was Alternaria alter-nata.The optimum culture conditions for the pathogen were 28℃,pH 9,and the optimum carbon and nitrogen sources for growth were starch and potassium nitrate,respectively.The EC50 of 1%osthole AS,0.5%physcion AS and 5%allicin ME were 19.772,243.245 and 1 810.685 mg/L,respectively.Conclusion:The pathogen of black spot disease of Paeonia lactiflora in Xinyang area is Alternaria al-ternata.The optimum culture condition of the pathogen is temperature 28℃,pH 9,and the optimum carbon and nitrogen sources are starch and potassium nitrate,respectively,1%osthole AS has a strong inhibitory effect on the pathogen.关键词
