基于DNA特征片段的驯鹿源性成分PCR检查方法研究
Study on PCR Detection Method of Rangifer tarandus-Derived Ingredients Based on DNA Characteristic Fragments

Abstract
Objective:To establish a PCR method for the detection of Cervi Cornu Pantotrichum common counterfeit Rangifer tar-andus-derived ingredients.Methods:According to the mitochondrial cytochrome oxidase I gene(COI)sequence of Cervi Cornu Pantotri-chum and its common counterfeit Rangifer tarandus,template DNA quality control primers and Rangifer tarandus-derived ingredients detection primers were designed,respectively,and the related factors affecting the PCR reaction system,such as annealing temperature,cycle times and template amount,were screened and optimized.Results:When the template DNA met the quality control requirements,specific bands with the size of 554 bp could be obtained.A specific band with a size of 237 bp could be obtained if Rangifer tarandus-derived ingredients were present in the sample,and no band could be detected at 237 bp if the sample was Cervi Cornu Pantotrichum from Cervus nippon or Cervus elaphus.The results of methodological investigation showed that the method had strong specificity,high de-tection rate,good sensitivity and stability for counterfeit products.Conclusion:The method for the examination of Cervi Cornu Pantotri-chum common counterfeit Rangifer tarandus-derived ingredients established in this research can be used to supplement the existing standard for the identification of the authenticity of the basic ingredients of Cervi Cornu Pantotrichum,and can quickly and accurately i-dentify whether it is Rangifer tarandus-derived ingredients.关键词
