Abstract

Objective:To establish the HPLC fingerprint and multi-component content determination method for the decoction pieces of Astragali Complanati Semen,and to evaluate the quality of Astragali Complanati Semen by combining with the chemical pattern recognition method.Methods:The fingerprint of 22 batches of Astragali Complanati Semen decoction pieces was established by HPLC gradient elution,and the similarity evaluation was carried out by using the"Similarity Evaluation System of Traditimal Chinese Medicine Chromatographic Fingerprint(2012 Edition)".Cluster analysis,principal component analysis and orthogonal partial least squares-dis-criminant analysis were performed using SPSS 23.0 and SIMCA 14.1 software to screen the differential components.The contents of seven flavonoid components were determined.Results:The fingerprint of 22 batches of Astragali Complanati Semen decoction pieces was labeled with 16 peaks,and the similarity was greater than 0.950.The samples could be classified into 4 groups according to the origin by cluster analysis.Nine differential markers were obtained by synthetic analysis.The contents of ampelopsin,astragalin,complanatoside B,complanatoside,quercetin,kaempferol and formononetin in the samples ranged from 2.2699~3.1909 mg/g,0.1473~0.2576 mg/g,0.1730~0.7083 mg/g,0.5028~0.7486 mg/g,0.0448~0.2523 mg/g,0.0266~0.0441 mg/g,0.0588~0.2380 mg/g,respectively.Conclusion:The HPLC fingerprint and multi-component content determination method established in this study has good stability and feasibility,and can provide a reference for the quality evaluation of Astragali Complanati Semen decoction pieces by combining with the chemical pattern recognition.
