苦参及其炮制品水煎液对α-萘异硫氰酸酯诱导的黄疸大鼠作用比较
Comparison of the Effect of Sophorae Flavescentis Radix and Its Different Processed Products Decoction on α-Naphthyl Isothiocyanate-Induced Jaundice in Rats

Abstract
Objective:To compare the therapeutic effects of Sophorae Flavescentis Radix and its different processed products de-coction on α-naphthyl isothiocyanate(ANIT)-induced jaundice in rats,so as to provide experimental evidence for the clinical use of dif-ferent processed products of Sophorae Flavescentis Radix.Methods:Seventy Wistar rats were randomly divided into normal control group,model group,raw Sophorae Flavescentis Radix group,Sophorae Flavescentis Radix processed with wine group,Sophorae Flaves-centis Radix steamed with wine group,stir-fried Sophorae Flavescentis Radix with bran group,Sophorae Flavescentis Radix steamed with swill group with 10 rats in each group.Each group was administered intragastrically at a dose of 10 mL/kg,once a day,for 9 consecutive days;The rats in normal control group and model group were given equal amounts of normal saline;on the 7th day,2 hours after the last administration,except for the normal control group,which was given an equal volume of olive oil by intragastric administration,the other groups were given ANIT olive oil solution(10 mg/mL)according to 60 mg/kg.On the 9th day of the experiment,2 hours after the last intragastric administration,blood from the abdominal aorta,liver and spleen tissues were extracted,and the pathological changes of liver tissues were observed under the microscope after HE staining.The levels of ALT,AST,ALP,TBiL and TBA in serum and the contents of NTCP,BSEP,MRP-2 and FXR in liver tissue were determined by ELISA.Results:Compared with model group,the levels of ALT,ALP,TBiL and TBA in serum and the content of BSEP in liver tissue were significantly decreased,while the contents of NTCP,MRP-2 and FXR in liver tissue were significantly increased in each administration group(P<0.05 or P<0.01).The indexes of rats in raw Sophorae Flavescentis Radix group were more significant than those of other Sophorae Flavescentis Radix processed product groups.Conclusion:Sophorae Flavescentis Radix and its different processed products have interventional therapeutic effects on ANIT-induced jaundice rats,and raw Sophorae Flavescentis Radix has a better effect.The mechanism may be related to the activation of FXR receptor,the regenera-tion of liver cells,the reduction of BSEP content in vivo,the activation of NTCP and MRP2 transporters,and the promotion of bile acid effection.关键词
