摘要

Abstract

Objective:To observe the inhibitory effect of Paridis Rhizoma saponins on HepG2 cells,and to further study its anti-hepatoma mechanism.Methods:HepG2 cells were used to verify the action mechanism of drug,cell proliferation capacity was detected by MTT,cell migration capacity was detected by nicks and Transwell tests,cell morphological changes were observed by inverted micro-scope,cell proliferation capacity was detected by colony formation assay,cell apoptosis was detected by flow cytometry,ROS production level was detected by fluorescence method.The expressions of apoptosis-related proteins and pathway proteins were detected by Western Blot.Results:Paridis Rhizoma saponins could decrease proliferation,clonogenesis and migration of HepG2 cells,increase apoptosis rate and ROS level,decrease the expressions of Bcl-2,PI3K,p-PI3K,AKT,p-AKT,JAK2,STAT3,p-STAT3,mTOR and p-mTOR proteins,increase the expressions of Bax,cleaved Caspase-3,PERK,ATF4 and CHOP proteins.Conclusion:Paridis Rhizoma saponins plays an anti-liver cancer role by inhibiting PI3K/AKT pathway and activating PERK/ATF4/CHOP mediated apoptosis.
