Abstract

[Objective]To improve the success of hybridization among Encore® azalea varieties and identify suitable hybrid parents.[Method]The experiment was conducted using three double-petal varieties of Encore® azalea were used as the maternal materials to carry out stigma pollination determination.At the same time,five single petal varieties were used as the male parents for pollen viability comparisons.Finally,the hybrid fruiting characteristics of 15 hybrid combinations were analyzed to comprehensively evaluate their hybrid compatibility.[Result]1)The stigmas of the double-petal varieties of Encore® azalea were strong acceptability in full bloom and at the end of the flowering period;2)Of the 3 staining methods,0.5%MTT staining is an effective method for rapid detection of pollen viability in Rhododendron,in the five male parents measured,'Fuchsia Parasol'had the highest pollen viability(81.07%),'Red Tiara'was next(75.61%),'Ivory'was in the middle at 63.87%,the'Orchid Showers'and'Royalty'were lower at 41.09%and 51.61%,respectively;3)The results of pollen germination in vitro method for detecting pollen viability in the parent showed that'Ivory'had the highest pollen viability,and the'Royalty'pollen did not germinate.Through orthogonal experiments,low concentrations of H3BO3 and CaCl2 stimulate pollen germination,while high concentrations of sucrose significantly promote pollen germination.Specifically,for'Ivory',the optimal medium consisted of 100 g·L-1 sucrose+200 mg·L-1 H3BO3+50 mg·L-1 CaCl2.In contrast,for'Red Tiara','Orchid Showers',and'Fuchsia Parasol',the preferred medium was 150 g·L-1 sucrose+100 mg·L-1 H3BO3+50 mg·L-1 CaCl2;4)Of the 15 hybrid combinations tested in this study,12 hybrid combinations were fertile.Based on the analysis of compatibility,the hybrid combinations using'Empress'as the female parent all exhibited strong compatibility,'Ivory'as the male parent displayed strong compatibility when hybridized with three female parents.However,'Royalty'showed extremely poor compatibility when hybridized with double-petaled varieties.[Conclusion]There are certain differences in the hybrid compatibility among different varieties of Encore® azalea.Among them,'Empress'is suitable as the female parent,and'Ivory'is suitable as the male parent.
