Analysis of ISSR Genetic Diversity of Polygala tenuifolia Germplasm Resources and Its Association with Active Components

Abstract
Objective:To study the genetic diversity of 36 Polygala tenuifolia germplasm from different sources and determine the content of active components,to screen the genetic loci related to active components,so as to provide the basis for variety selection of Polygala tenuifolia.Methods:ISSR marker was used to analyze genetic diversity and population genetic structure of 36 Polygala tenuifo-lia,and MLM method was used to analyze the correlation between the marked results and active components.Results:There was a cer-tain genetic diversity among the 36 Polygala tenuifolia germplasm,and the genetic distance ranged from 0.1335 to 0.6131.According to UPGMA clustering analysis,36 Polygala tenuifolia germplasm could be divided into two categories at 0.63.The average contents of polygalaxanthone Ⅲ,3,6′-disinapoylsucrose and tenuifolin in Polygala tenuifolia from different sources were 0.15%,0.61%,2.30%,respectively.36 Polygala tenuifolia germplasm were divided into three subpopulations by population genetic structure analysis.The above results were used to analyze the MLM model,which showed that there were 6 primers and 13 loci associated with the three active components under significant association(P<0.05),and the interpretation rate ranged from 11.69%to 29.63%.Conclusion:The germ-plasm resources of Polygala tenuifolia have high genetic variability and gene flow,and the developed ISSR loci can provide basis for high quality molecular breeding.关键词
