Abstract

Objective:To establish a particle-specific polymerase chain reaction(PCR)method for the identification of Cicadae Periostracum formulation granules,to effectively realize the identification of Cicadae Periostracum inmedicinal materials and different dosage forms,so as to ensure the safety and effectiveness of clinical medication.Methods:Based on the Cryptotympana pustulata cyto-chrome oxidaseⅠ(COⅠ)gene sequence,single nucleotide polymorphism(SNP)sites were screened,specific primers CT-F and CT-R were designed,and a specific PCR method was established for the identification of Cicadae Periostracum formulation granules.The main conditions affecting PCR reaction were optimized,and the durability and applicability of the method were investigated.Results:When the annealing temperature was 57℃and the number of cycles was 37 times,both the Cicadae Periostracum and its formulation granules showed a single bright band at 173 bp,while the counterfeit had no band.Conclusion:The established specific PCR identification meth-od can accurately identify Cicadae Periostracum,standard decoction freeze-dried powder and formulation granules,and also provide a reference for the quality standard research of Cicadae Periostracum formulation granules.
