基于TNF-α/TNFR1/TRADD信号通路探讨定痫丸对癫痫小鼠海马神经元凋亡的影响
Effect of Dingxian Pills on Hippocampal Neuron Apoptosis in Epileptic Mice Based on TNF-α/TNFR1/TRADD Signaling Pathway

Abstract
Objective:To investigate the effect of Dingxian Pills on hippocampal neuron apoptosis in epileptic mice based on TNF-α/TNFR1/TRADD pathway.Methods:60 Kunming mice were randomly divided into normal control group,model group,carbamazepine(30 mg/kg)positive control group and Dingxian Pills low(4.08 g/kg),medium(8.16 g/kg)and high(16.32 g/kg)dose groups with 10 in each group.The epileptic mouse model was established by pilocarpine induction,and the mice were given continuous administra-tion of the drug for 2 weeks,once a day,and the success of the model was observed by eleetroencephalogram.The pathological changes of hippocampus were observed by HE staining,hippocampal neuron apoptosis was observed by TUNEL staining,the expressions of TNF-α,TNFR1 and TRADD mRNA and proteins in hippocampus were detected by immunohistochemical method(IHC),Real-time PCR,Western Blot.Results:Compared with the normal control group,eleetroencephalogram continuous epileptic radio wave in model group was increased,hippocampal neuron morphological structure was damaged,hippocampal neuron apoptosis was aggravated,and TNF-α,TNFR1,TRADD mRNA and protein expressions in hippocampus were significantly increased(P<0.05).Compared with model group,eleetroencephalogram epileptic discharge was reduced,hippocampal neuron morphological structure was significantly recovered,hipp-ocampal neuron apoptosis was improved to varying degrees,and the expressions of TNF-α,TNFR1 and TRADD mRNA and protein in hippocampus were significantly decreased in each administration group(P<0.05).Conclusion:Dingxian Pills can alleviate the apoptosis of hippocampal neurons by inhibiting TNF-α/TNFR1/TRADD signaling pathway,and finally play an anti-epileptic role.关键词
