基于PPAR-γ和TLR-4/NF-κB信号通路研究黄花倒水莲多糖对非酒精性脂肪肝大鼠的影响

Abstract
Objective:To evaluate the effect of Polygala fallax polysaccharide on nonalcoholic fatty liver disease(NAFLD)model rats and explore its possible mechanism.Methods:60 SD rats were randomly divided into normal control group,model group,silymarin group and low,medium and high dose of Polygala fallax polysaccharide groups.Except the normal control group,the rats in other groups were fed high-fat and high-sugar diet for 14 weeks to form NAFLD model.From the 9th week,the rats in each administration group were given the corresponding drug intragastric treatment for 6 weeks.After the last intragastric infusion,water was not allowed for 16 hours,followed by eye blood collection and liver sampling.The pathological changes of rat liver tissue were observed by HE staining.The levels of serum ALT,AST,TG,TC,LDL-C,HDL-C and liver SOD,GSH-Px and MDA were detected by biochemical method.The levels of IL-6,IL-1β and TNF-α in liver tissue were detected by ELISA.The PPAR-γ,TLR-4,P-NF-κB p65 protein expressions in liver tissue were detected by Western Blot.Results:Compared with the normal control group,serum ALT,AST,TG,TC,LDL-C levels in model group were significantly increased,HDL-C levels were significantly decreased,SOD,GSH-Px levels and PPAR-γ protein expression in liver tissue were significantly decreased.The levels of MDA,IL-6,IL-1β,TNF-α and the protein expressions of TLR-4 and P-NF-κB p65 were significantly increased(P<0.01).Compared with model group,serum ALT,AST,TG,TC and LDL-C levels in silymarin group and Po-lygala fallax polysaccharide medium and high dose groups were significantly decreased,HDL-C level was significantly increased,SOD and GSH-Px levels and PPAR-γ protein expression in liver tissue were significantly increased.MDA,IL-6,IL-1β,TNF-α levels and TLR-4,P-NF-κB p65 protein expressions were significantly decreased(P<0.05 or P<0.01).Conclusion:Polygala fallax polysaccha-ride has a good protective effect on NAFLD model rats,and the mechanism may be related to the inhibition of PPAR-γ,TLR-4/NF-κB pathway signal transduction.关键词
