黄芪苷甲Ⅳ协同人胎盘间充质干细胞移植对卵巢功能不全大鼠卵巢功能的保护作用
Protective Effect of Astragaloside Ⅳ and Human Placental Mesenchymal Stem Cell Transplantation on Ovarian Function in Rats with Ovarian Dysfunction

Abstract
Objective:To investigate the effect of astragaloside Ⅳ on the hPMSCs transplantation to treat POI rats.Methods:50 SD rats were randomly divided into normal control group,model group,astragaloside Ⅳ group,hPMSCs group,astragaloside Ⅳ with hPM-SCs group,with 10 rats in each group.The POI rat model was established after continuous intraperitoneal injection of cyclophosphamide(CTX)at 50 mg/kg concentration for 15 days.The rats in Astragaloside Ⅳ group and astragaloside Ⅳ with hPMSCs group were given astragaloside Ⅳ(40 mg/kg)from the 10th day of modeling.The rats in hPMSCs group and Astragaloside Ⅳ with hPMSCs group were injected with 2.5×106 hPMSCs in the tail vein on the 15th and 22nd day of modeling,respectively.The levels of sex hormones,antioxi-dant enzymes and pro-inflammatory factors in serum were detected by ELISA,ovarian tissue structure was observed by HE staining,ovarian cell apoptosis was detected by TUNEL method,the expressions of ovarian functional markers and apoptotic markers were detec-ted by Western Blot and immunohistochemical methods.Results:The estrous cycle of rats was disturbed after CTX induction,and the POI model was established successfully.Compared with the model group,the levels of serum FSH,IL-6,IL-1β and TNF-α were de-creased,AMH,E2,SOD and GSH-Px levels were increased,the ovarian histopathology was significantly improved,the apoptosis of ovari-an cells was decreased,and the expressions of C/EBP-β,ERK and Bcl-2 proteins in ovarian tissue were increased,Bax,cleaved Caspase-3 protein expressions were decreased.The effect of astragaloside Ⅳ with hPMSCs is better than that of astragaloside Ⅳ and hPMSCs transplantation.Conclusion:Astragaloside Ⅳ has antioxidant and anti-inflammatory effects,and can enhance the effect of hPM-SCs transplantation in the treatment of POI by improving the microenvironment in the injured ovarian tissue.关键词
