基于TNF-α诱导的MH7A细胞模型研究附子总生物碱与白芍总苷配伍治疗类风湿关节炎的协同机制
Synergistic Effects of Aconiti Lateralis Radix Praeparaia Total Alkaloids and Paeoniae Radix Alba Total Glycosides in the Treatment of Rheumatoid Arthritis Based on TNF-α-Induced MH7A Cell Model

Abstract
Objective:To explore the potential molecular mechanism of the combination of Aconiti Lateralis Radix Praeparaia total alkaloids and Paeoniae Radix Alba total glycosides,the main active components of Aconiti Lateralis Radix Praeparaia and Paeoniae Ra-dix Alba,used in traditional Chinese medicine for the treatment of rheumatoid arthritis.Methods:CCK-8 method was used to screen the safe dose of Aconiti Lateralis Radix Praeparaia total alkaloids to MH7A cell model.Western Blot,immunofluorescence and ELISA were used to analyze the effects of Aconiti Lateralis Radix Praeparaia total alkaloids and Paeoniae Radix Alba total glycosides on the expres-sion of NF-κB signaling pathway protein and some downstream cytokines,and their anti-inflammatory effects were evaluated.Western Blot and flow cytometry were used to analyze the effects of Aconiti Lateralis Radix Praeparaia total alkaloids and Paeoniae Radix Alba total glycosides on mitochondrial apoptosis signaling pathway protein expression,apoptosis rate and cell cycle.Results:Compared with model group,Aconiti Lateralis Radix Praeparaia total alkaloids had inhibitory effect on cell proliferation.The combination of Aconiti Lateralis Radix Praeparaia total alkaloids and Paeoniae Radix Alba total glycosides could inhibit the production of IL-1β,IL-6,MMP-1,MMP-3,PGE2,TGF-β and VEGF by reducing the protein expressions of p-IKBα,p-IKKβ and P65.By increasing the expressions of Bax and Caspase-3 proteins,decreasing the expression of Bcl-2 protein,the apoptosis was promoted.The cell cycle was blocked and re-mained in the G0/G1 phase.Conclusion:Aconiti Lateralis Radix Praeparaia total alkaloids and Paeoniae Radix Alba total glycosides ex-ert anti-inflammatory and apoptotic effects by acting on NF-κB and mitochondrial apoptotic signaling pathway,and also participate in cell cycle regulation and inhibit cell proliferation.The synergistic effect of Aconiti Lateralis Radix Praeparaia total alkaloids and Paeoni-ae Radix Alba total glycosides against rheumatoid arthritis may be related to partial overlap of therapeutic targets.关键词
