Study on the Protective Mechanism of Bicuculline and Tetrahydrocoptisine Against H2O2-Induced Cardiomyocyte Injury

Abstract
Objective:To investigate the protective effect of Bicuculline and tetrahydrocoptisine on H2O2-induced H9c2 cell injury and its mechanism.Methods:H9c2 cells were divided into normal control group,model group,N-acetyl-L-cysteine(NAC)positive con-trol group,Bicuculline(100 μmol/L)group and tetrahydrohydrazine(100 μmol/L)group.The H9c2 cell oxidative injury model was in-duced by H2O2.After the intervention,the survival rate of cardiomyocytes was detected by CCK-8 method,the cell morphology was ob-served,and the release of free oxygen(ROS),mitochondrial membrane potential and apoptosis of cardiomyocytes in each group were de-tected.The levels of lactate dehydrogenase(LDH)and malondialdehyde(MDA)in cell culture supernatant and the activities of super-oxide dismutase(SOD),catalase(CAT)and glutathione peroxidase(GSH-Px)in cells of each group were detected.The expressions of apoptosis-related proteins Bcl-2,Bax,Caspase-3 and Caspase-9 were detected.Results:Compared with the normal control group,myo-cytes in the model group showed deformation,void enlargement and death,cell survival rate was significantly decreased,apoptosis rate,ROS level,mitochondrial membrane potential and LDH and MDA levels in culture supernitant were significantly increased,while SOD,CAT,GSH-Px levels and Bcl-2 protein were significantly decreased.The protein expressions of Bax,Caspase-3 and Caspase-9 were sig-nificantly increased(P<0.01).Compared with model group,cell morphology in each administration group was close to normal,cell sur-vival rate was significantly increased,ROS level,apoptosis rate,mitochondrial membrane potential and LDH and MDA levels in culture supernants were significantly decreased,and SOD,CAT and GSH-Px levels were significantly increased.The protein expressions of Bax and Caspase-9 were significantly decreased(P<0.01).Conclusion:Bicuculline and tetrahydrocoptisine have a good protective effect on H2O2-induced H9c2 cell damage,and the mechanism is related to reducing cell oxidative damage,inhibiting cell apoptosis and stabili-zing mitochondrial membrane potential.关键词
