摘要

Abstract

Sheep infected with Sarcocystis gigantea will have symptoms such as vomiting and enteritis,which will bring huge economic losses to the livestock industry.In this study,the whole protein of Sarco-cystis gigantea was used as the antigen to optimize the reaction conditions,and an indirect ELISA method for the detection of Sarcocystis gigantea was established with short time,low cost and can be operated be-fore the animal's death.The established ELISA method was used to detect Sarcocystis gigantea in 538 Ti-betan sheep serum samples from 6 regions of Qinghai province.The results showed that the coating concen-tration was 0.05 μg,the serum dilution ratio was 1∶100,and the secondary antibody dilution ratio was 1∶4 000.The positive rates of serum IgG and IgM in Tibetan sheep in six regions of Qinghai province were 16.2%and 20.3%,respectively.The results showed that an indirect ELISA method was successfully estab-lished using the whole protein of Sarcocystis gigantea,which had good reproducibility and could be used for the preliminary detection of Sarcocystis gigantea infection in Tibetan sheep.
