两种不同佐剂猪流感灭活疫苗小鼠皮内免疫效果比较
Comparison of Intradermal Immune Effect of Swine Influenza Virus Inactivated Vaccines with Two Different Adjuvants

Abstract
Intradermal immunity is an efficient immune route for vaccine administration.There is an urgent need of safe and efficient adjuvants for intradermal immunity.In this study,commercial adjuvants Gel02 and ISA15 were used to develop swine influenza virus(SIV)inactivated vaccines,and the local safety of skin and immune efficacy were evaluated in Balb/c mice.The local reactions of skin were examined from 0-8 d,then the antibodies in serum and bronchoalveolar lavage fluid(BALF)were detected on 28d.The re-sults indicated that neither of the adjuvants caused significant local skin reactions,such as redness,swelling and breakage.Meanwhile,Gel02 and ISA15 both induced temporary up-regulation of inflammatory cyto-kines and aggregation of inflammatory cells in the skin.In addition,Gel02 was completely absorbed as early as 24h,which was faster than ISA15.Hemagglutination inhibition(HAI)and enzyme linked immunosor-bent assay(ELISA)results indicated that ISA15 and Gel02 adjuvanted influenza vaccines induced higher level of antibodies compared to H3N2 alone.Gel02 tended to elicit IgG2c and IgG2a antibodies.However,ISA15 tended to elicit IgG1 antibodies.In BALF,Gel02 adjuvanted vaccine induced higher level of IgG anti-bodies,but not IgA antibodies.However,ISA15 could not induce the expression of IgG and IgA antibodies in BALF.In summary,the intradermal immune efficacy and safety of Gel02 and ISA15 were first confirmed in this study,which indicated that Gel02 was more suitable for intradermal immunity of SIV inactivated vaccines.This study established an important foundation for the development of swine influenza intradermal vaccines in the future.关键词
