基于磁共振波谱技术探讨柴胡疏肝散对围绝经期肝郁证大鼠认知功能的影响
Effect of Chaihu Shugan Powder on Cognitive Function in Perimenopausal Rats with Liver-Stagnation Syndrome Based on Magnetic Resonance Spectroscopy

Abstract
Objective:To investigate the effect of Chaihu Shugan powder on cognitive function of perimenopausal rats with liver-stagnation syndrome based on magnetic resonance spectroscopy.Methods:8-month-old female SD rats were fed normally until 12 months of age,and were divided into control group,model group and intervention group using the random number table method.The con-trol group was not given any stimulation and only normal grasping was performed;the model and intervention groups were used chronic unpredictable mild stress method(CUMS)to establish a perimenopausal liver-depression syndrome model,which lasted for 8 weeks.In the sixth week of modeling,the intervention group was given Chaihu Shugan powder by gavage at a dose of 0.331 g/(kg·d),while the con-trol and model groups were given sterile physiological saline by gavage at a dose of 10 mL/(kg·d),once a day for three consecutive weeks of intervention.The sugar water preference experiment and forced swimming experiment were used to detect the sugar water preference rate and forced swimming immobility time to evaluate depression like behavior in rats.The new object recognition experiment and Morris water maze experiment were used to detect the new object recognition index,escape latency,and platform crossing frequency to assess the cognitive and memory abilities of rats.The small animal MRI machine was used to collect signals of N-acetyl-aspartate(NAA),gluta-mate(Glu),myo-inositol(mI),and creatine(Cr)in the left hippocampus of the brain,and calculate their peak areas.Using the Cr value as a reference standard,the ratio of the NAA,Glu,and mI peak areas to Cr was calculated.Results:1)Depression like behavior evalua-tion:Compared with the control group,the model group showed a significant decrease in sugar water preference rate(P<0.05)and a sig-nificant prolongation of forced swimming immobility time(P<0.05).Compared with the model group,the intervention group showed a sig-nificant increase in sugar water preference rate(P<0.05)and a significant reduction in forced swimming immobility time(P<0.05).2)Evaluation of cognitive and memory abilities:Compared with the control group,the model group showed a significantly prolonged escape latency(P<0.05),and a significant decrease in new object recognition index and platform crossing frequency(P<0.05).Compared with the model group,the intervention group showed a significant reduction in escape latency(P<0.05),and a significant increase in new object recognition index and platform crossing frequency(P<0.05).3)Neurometabolite level detection:Compared with the control group,the model group showed a significant reduction in NAA/Cr and Glu/Cr of the left hippocampus(P<0.05),and a significant increase in mI/Cr(P<0.05).Compared with the model group,the intervention group showed a significant increase in NAA/Cr and Glu/Cr(P<0.05),and a significant decrease in mI/Cr(P<0.05).Conclusion:The evidence of liver depression in perimenopausal rats may be closely related to the alteration of neurometabolites in the hippocampal brain region,and Chaihu Shuohe powder may improve the cognitive impairment in-duced by liver depression in perimenopausal period by regulating the hippocampal levels of NAA,Glu and mI.关键词
