Simultaneous Determination of 5 Components in Huazhuo Sanjie Chubi Fang by HPLC

Abstract
Objective:To establish a high performance liquid chromatography(HPLC)method for the preliminary determination of phelloberine hydrochloride,isozipidine,astibin,berberine hydrochloride and rosaminic acid in Huazhuo Sanjie Chubi Fang,and to provide experimental basis for the quality control of prescription in clinical use and subsequent improvement of dosage form.Methods:The analysis was performed on a 45℃thermostatic Phenomenex Geminic C18 column(4.6 mm×250 mm,5 μm),with the mobile phase comprising of acetonitrile-0.2%phosphoric acid flowing at 0.9 mL/min,and detection wavelengths were set at 206,206,290,264,330 nm.Results:The linear ranges of phelloberine hydrochloride,isozipidine,astilbin,berberine hydrochloride and rosa-minic acid were 0.60-59.85,0.61-61.20,2.42-242.40,1.43-142.80 and 0.74-74.40 μg/mL,and showed a good linear relationship within their respective ranges(r≥0.999 8).The RSD of precision,repeatability and stability were all less than 2.1%,and the average recovery rate was 97.4%～104.9%,RSD≤2.20%.The average content of phelloberine hydrochloride,isozipidine,astilbin,berberine hydrochloride and rosaminic acid were 96.22,51.89,771.75,220.40 and 202.72 μg/g,respectively.Conclusion:The method is sensi-tive,accurate,repeatable and stable.It is suitable for the quality evaluation and quality control of Huazhuo Sanjie Chubi Fang.关键词
