摘要

Abstract

Objective:To study the effect of Taishan Panshi San(TSPSS)on oxidative stress(OS)in placental tissue of spontane-ous abortion(SA)model rats.Methods:A total of 18 female and 18 male SD rats were cohabitated overnight at a 1∶1 ratio for mating.The day of vaginal plug detection was designated as the first day of pregnancy.18 pregnant rats were divided into normal group,model group,and treatment group,with 6 rats in each group,using a random number table method.On the 1st to 11th day of preg-nancy,the treatment group was given TSPSS solution by gavage at a dose of 10 g/(kg∙d),while the normal and model groups were given physiological saline solution by gavage at a dose of 10 mL/(kg∙d).On the 6th to 8th day of pregnancy,the model and treatment groups were subcutaneously injected with 0.4 mg/(kg∙d)of bromocriptine solution,while the normal group was subcutaneously in-jected with 1 mL/(kg∙d)of 75%ethanol solution.The body mass of rats was measured every 2 days during pregnancy.The embryo morphology is observed and the embryo absorption rate is calculated;HE staining was used to observe the pathological changes of maternal-fetal interface placental tissues;the serum level of 8-iso-prostaglandin F2α(8-iso-PGF2α)was detected by ELISA;the levels of glutathione peroxidase(GSH-Px)and malondialdehyde(MDA)in placental tissues and serum were detected;the mRNA and pro-tein expression levels of superoxide dismutase(SOD)and quinone oxidoreductase-1(NQO-1)in placental tissues were detected by qPCR and Western blot.Results:The observation of uterine embryo morphology showed that the embryos of the normal group exhib-ited a bead-like arrangement with pale pink or red coloration;the embryos of the model group appeared dark purple,with severe con-gestion and a significant reduction in the volume of some embryos;the embryos of the treatment group coloration returned to pale pink/red,with markedly alleviated congestion,closely resembling the normal group.The HE staining results showed that the arrange-ment of cells on the placental side of the maternal-fetal interface in the model group was irregular compared to the normal group,and the blood vessels were severely dilated;the arrangement of cells on the placental side of the maternal-fetal interface in the treatment group was more regular than that in the model group,and vascular dilation was significantly improved.Compared with the normal group,the body mass of the model group decreased significantly on the 9th and 11th day of pregnancy(P<0.05);the embryo absorp-tion rate increased significantly(P<0.05);the levels of serum 8-iso-PGF2α,MDA,and placental tissues MDA significantly increased(P<0.05),while the levels of GSH-Px in serum and placental tissues significantly decreased(P<0.05);the mRNA and protein expres-sion of SOD and NQO-1 in placental tissues significantly reduced(P<0.05).Compared with the model group,the embryo absorption rate of the treatment group significantly reduced(P<0.05);the levels of serum 8-isoPGF2α,MDA,and placental tissues MDA sig-nificantly reduced(P<0.05),and the content of GSH-Px in serum and placental tissues significantly increased(P<0.05);the mRNA and protein expression of SOD and NQO-1 in placental tissues significantly increased(P<0.05).Conclusions:TSPSS may achieve fe-tal protection by promoting the Nrf2 antioxidant pathway,inhibiting oxidative stress in placental tissue,and reducing placental tissue damage.
