Abstract

A liquid chromatography-tandem mass spectrometry method has been established for the determination of asparagine and glutamine in special dietary foods.Following the process of direct ex-traction or enzymatic extraction,the internal standard was added and the sample was separated by po-lar C18 chromatographic column(150 mm×4.6 mm,2.6 μm).The mobile phases consisted of aceto-nitrile and 0.02%formic acid in water.It was detected in a positive ion mode of electric spray for quantitative measurement and qualitative check.The results showed that there was a good linear rela-tionship between asparagine and glutamine with correlation coefficients greater than 0.999.The lin-ear range was 0-1 000 ng/mL.The limits of detection(LODs)were 0.8 mg/100g and 4.0 mg/100g for liquid and solid samples,respectively,and the limits of quantification(LOQs)were 2.4 mg/100g and 12 mg/100g for liquid and solid samples,respectively.The recoveries of the method ranged from 83.5%to 104%,with relative standard deviations ranging from 0.28%to 4.2%(n=6).This method has the advantages of simple pre-treatment,short analysis time,good accuracy,and high practicali-ty.It can be used to precise quantification of asparagine and glutamine in specialty foods.
