目的:在磷灰石-硅灰石(Apatite-wollastonite,AW)生物活性玻璃陶瓷中分别引入Sr、La、Co等元素,研究其对骨修复材料的影响.方法:利用溶胶-凝胶法,在AW生物活性玻璃陶瓷体系中分别掺入Sr、La、Co等元素,制成Sr-AW、La-AW、Co-AW骨修复材料;利用X射线衍射、傅里叶变换红外光谱、Tris-HCl体外浸泡降解实验、CCK-8细胞活性实验和细胞活/死染色实验对材料的理化特性和细胞相容性进行初步探索.结果:少量Sr或La或Co(1.8 mol%)元素的掺杂均不会改变材料的主晶相,当掺杂量达到7.2 mol%时Sr-AW出现了新的硅酸锶钙(CaSrSiO4,PDF#77-1619)晶相,而La-AW的主晶相则变为磷酸镧(LaPO4,PDF#73-0188).Sr元素明显加快了材料降解,Co和La对材料的降解也有一定的促进.高浓度的Co和La元素有较强的细胞毒性,而Sr元素在掺杂量为7.2 mol%时仍有较好的细胞相容性.结论:在AW中掺入合适浓度的Sr等元素可以调控材料的降解速率,高浓度Co或La的掺杂在体外细胞实验中产生了一定的毒性,而同样浓度的Sr-AW则无显著的细胞毒性.

Objective:In this study,Sr,La and Co were introduced into apatite-wollastonite(AW)bioactive glass-ceramic to modify the properties of bone repair materials.Methods:Sr,La and Co were added into AW bioactive glass-ceramic by sol-gel method,respectively.The physical and chemical properties and cell compatibility of the materials were assessed by X-ray diffraction,Fourier transform infrared spectroscopy,in vitro degradation experiment,CCK-8 assay and live/dead cell staining experiment.Results:The results showed that incorporation of 1.8 mol%amount of each element into the material yielded no significant differences in terms of the main crystal phase compared with AW.But when the doping amount reaches 7.2 mol%,a new crystal phase(CaSrSiO4,PDF#77-1619)appears in Sr-AW,while the main crystal phase of La-AW changes to LaPO4(PDF#73-0188).The degradation rate of Sr-AW was greatly accelerated when 7.2 mol%Sr were doped to AW.Meanwhile the dissolution rate of Co-AW and La-AW were also accelerated.Cellular experiments showed that the incorporation of 7.2 mol%of Co or La was cytotoxic in vitro,while the Sr-AW still had good cytocompatibility at the same doping concentration.Conclusion:The appropriate concentration of Sr in AW can regulate the degradation rate of materials.High concentration of Co or La doping has a certain cellular toxicity,while the same concentration of Sr-AW has no significant effect on cell viability.
