Abstract
As an important member of the N6-methyladenosine(m6A)methyltransferase family,methyltransferase like 16(METTL16)dynamically regulates tumorigenesis and development through epigenetic modification.Although the importance of METTL16 in various tumors has been confirmed,there is still a lack of systematic summary and analysis of its molecular mechanism heterogeneity and functional diversity in different tumor types.Based on the latest research progress,this article comprehensively reviews the molecular mechanism of METTL16 in tumorigenesis and development and its clinical transformation value.Studies have shown that METTL16 can requlate the downstream target gene network through m6A-dependent and m6A-independent mechanisms,thereby affecting key biological processes such as tumor cell cycle progression,metabolic reprogramming and ferroptosis resistance.Clinical transformation studies have shown that the expression level of METTL16 is significantly associated with the prognosis of some tumors such as gastric cancer,hepatocellular carcinoma,and colorectal cancer.In terms of treatment strategies,interventions targeting METTL16 have made initial progress,including enhancing the therapeutic effect of pancreatic ductal adenocarcinoma by sensitizing poly adp-ribose polymerase inhibitors(PARPi),and improving the sensitivity of bladder cancer to cisplatin chemotherapy.These results not only reveal the important value of METTL16 as a key target for tumor therapy,but also provide a theoretical basis for the development of new anti-tumor strategies.关键词
