Polo样激酶1在肿瘤与炎症中生物学功能研究进展
Research progress on versatile biological functions of polo-like kinase 1 in tumors and inflammation
摘要
Abstract
Polo-like kinase 1(PLK1)is a serine/threonine protein widely present in eukaryotic cells. It plays a crucial role in the progression and regulation of the cell cycle.PLK1 is involved in physiological processes such as cell division, proliferation,DNA damage repair, and apoptosis. The aberrant overexpression of PLK1 in numerous tumors is significantly associated with rapid tumor cell proliferation and adverse prognosis, thereby positioning PLK1 as a promising therapeutic target for cancer treatment. In addition,PLK1 regulates cellular metabolic pathways such as the pentose phosphate pathway, aerobic glycolysis, and cholesterol metabolism. This not only promotes adaptive changes in normal cells but also provides energy for the proliferation of tumor cells.Emerging evidence highlights the crucial role of PLK1 in inflammatory responses, particularly during the early stages of inflammation.PLK1 is involved in the regulation of pyroptosis and modulates inflammatory responses by influencing the migration of immune cells, thus presenting novel therapeutic targets for inflammatory and autoimmune diseases. This review comprehensively elucidates the roles of PLK1 in regulating the cell cycle, modulating cell proliferation and apoptosis, regulating cellular energy metabolism, and participating in inflammatory responses. It aims to thoroughly investigate the multifaceted functions of PLK1 in cellular physiological and pathological processes, thereby providing a robust theoretical foundation for the development of innovative anti-tumor and anti-inflam-matory therapeutic strategies.关键词
