纳米二氧化钛(TiO2-NPs)对施氏鲟幼鱼的毒性效应研究
Toxic Effects of Nano-titanium Dioxide(TiO2-NPs)on Juvenile Acipenser schrenckii

Abstract
[Objective]The toxic effects of nano-titanium dioxide(TiO2-NPs)on the benthic fish Acipenser schrenckii were explored to provide the theoretic basis for evaluating the damage of TiO2-NPs to fish and its toxic effects.[Method]Taking juvenile A.schrenckii as the research material,96 h semi-lethal experiments(LC50)was conducted based on different TiO2-NPs concentrations of 50 mg/L,100 mg/L,200 mg/L,300 mg/L,400 mg/L,500 mg/L,600 mg/L and 800 mg/L.Based on the obtained data of the semi-lethal concentration of TiO2-NPs,three concentration gradients of TiO2-NPs(10%LC50,1%LC50,1‰ LC50)were set up respectively for the TiO2-NPs stress experiment by taking ultrapure water as control.The effects of TiO2-NPs on gills,liver,intestine and spleen of juvenile A.schrenckii were explored through microscopic observation,and the toxic effects of TiO2-NPs on juvenile A.schrenckii were clarified.[Result]The semi-lethal concentration tests showed that when the concentration of TiO2-NPs was 50 mg/L,the juvenile A.schrenckii began to die on the third day,with a mortality rate of 4.17%.When the concentrations were 100 mg/L,200 mg/L and 300 mg/L,the juvenile A.schrenckii began to die on the second day,and the mortality rates were 5.17%,8.33%and 12.5%respectively.When the concentrations were 400 mg/L,500 mg/L,600 mg/L and 800 mg/L,the juvenile A.schrenckii began to die on the first day,and the mortality rates were 8.33%,12.5%,16.67%and 20.83%respectively.The 96-hour LC50 of TiO2-NPs for juvenile A.schrenckii was 52.70 mg/L.High concentrations(10%LC50=5.27 mg/L)of TiO2-NPs damaged the morphological changes of the gills,spleen,liver and intestinal tissues of juvenile A.schrenckii,damaging the normal cell structures.The TiO2-NPs stress test showed that following 14-day exposure to TiO2-NPs,SOD activity of juvenile fish liver tissues increased with decreasing TiO2-NPs concentrations,CAT activity showed a gradual decline as TiO2-NPs concentrations reduced.GPx activity was significantly suppressed by high-concentration TiO2-NPs and MDA content demonstrated a downward trend with lower TiO2-NPs concentrations.[Conclusion]The 96-hour semi-lethal concentration of TiO2-NPs for juvenile A.schrenckii is 52.70 mg/L.TiO2-NPs can damage the cell structures of the gills,spleen,liver and intestinal tissues of juvenile A.schrenckii,causing oxidative stress response in liver tissue,and have obvious toxic effects on A.schrenckii.关键词
