紫锥菊多倍体母株子代的染色体变化分析
Analysis of Chromosomal Changes in the Progeny of Polyploid Mother Plants of Echi-nacea purpurea

Abstract
[Objective]This study aimed to explore the germination of seeds from tetraploid mother plants and the chromosomal changes in the natural progeny of tetraploid and triploid Echina-cea purpurea,and obtain aneuploids and meiotic tetraploid plants,so as to lay a solid foundation for the genetic research and variety improvement of Echinacea purpurea.[Method]Seed germina-tion experiments were carried out using the indoor germination method and the tissue-culture sterile method to determine the effects of these two different germination methods on the germination rates of the seeds from the tetraploid maternal plants and diploid maternal plants of the same strain of Echinacea purpurea.The root-tip chromosome counting method was employed to identify the ploi-dy of the offspring's chromosomes.[Result]The germination rate of Echinacea purpurea tetraploid seeds under the indoor germination method was 56.5%,which was 222.9%higher than that under the tissue-culture sterile germination method.Evidently,the indoor germination method is superior to the tissue-culture sterile germination method.In the identified progeny plants of tetraploid mother plants,for line 1,the chromosome number ranged from 30 to 35,39,42,44,with a total of 9 dif-ferent chromosome numbers.Fifty two triploid plants(3x=33)were obtained,accounting for 71.23%.For line 2,the chromosome number ranged from 31 to 34,43,with a total of 5 different chromosome numbers.Forty six triploid plants(3x=33)were obtained,accounting for 63.01%.Among the identified progeny plants of triploid mother plants,the chromosome number ranged from 21 to 26,with a total of 6 different chromosome numbers.The chromosome number of diploid plants(2x=22)obtained was 5,accounting for 17.86%.Among the progeny of triploid mother plants,the aneuploids with 23 chromosomes were the most common,accounting for 32.14%.[Con-clusion]The conventional tissue-culture sterilization method led to a decrease in the germination rate of tetraploid seeds.Significant variations were observed in the number of aneuploid chromosomes in the progenies derived from tetraploid Echinacea purpurea mother plants with distinct genotypes.New aneuploid materials such as monosomics and trisomics appeared in the progeny of triploid mother plants of Echinacea purpurea.These materials have a great value in genetic improvement and genetic theory research.关键词
