Abstract

Objective To explore theanti-inflammatory effects of Modified Heibu Gao(加味黑布膏)on a rat model of auricular composite acne.Methods 48 SPF-grade Wistar rats were randomly divided into six groups using a random number table:a blank group,a model group,a positive control group,and low,medium,and high dose groups of Modified Heibu Gao,with 8 rats in each group.Except for the blank group,the other groups were in-duced to develop a composite acne model by applying 100%oleic acid combined with an injection of Propionibacte-rium acnes to the auricular endothelium.After 3 weeks of continuous modeling,each drug group was treated with the corresponding medication,while the blank and model groups were treated with an equal amount of the base of Modi-fied Heibu Gao.After 4 weeks of continuous treatment,the severity of skin lesions on the auricular endothelium of each group was observed and scored.The levels of interleukin(IL)-1,IL-17,IL-23,and tumor necrosis factor-α(TNF-α)in the auricular endothelial lesion tissues were measured using enzyme-linked immunosorbent assay(ELISA).The gene expression levels of IL-1,IL-17,IL-23,and TNF-α in the auricular endothelial lesion tissues were detected using real-time fluorescent quantitative polymerase chain reaction(RT-PCR).Results Compared with the blank group,the model group showed significant thickening,redness,and desquamation of the auricular en-dothelial skin.The severity score of skin lesions,as well as the levels and gene expression of inflammatory cytokines IL-1,IL-17,IL-23,and TNF-α in the lesion tissues,were significantly increased(P<0.05).Compared with the mo-del group,the drug-treated groups showed significant improvement in skin swelling,thickening,desquamation,capil-lary dilation,and scab formation.The severity score of skin lesions,as well as the levels and gene expression of in-flammatory cytokines IL-1,IL-17,IL-23,and TNF-α in the lesion tissues,were significantly reduced(P<0.05).The high dose group of Modified Heibu Gao showed significantly better improvement in all indices compared to the low and medium dose groups(P<0.05).Conclusion Modified Heibu Gao exhibits potent anti-inflammatory effects on the auricular composite acne model in rats,significantly reducing the levels of inflammatory cytokines and their gene expression in skin lesion tissues,and improving symptoms such as skin thickening,redness,and desquamation.
