小苞黄芪多糖的分离、结构表征及抗氧化活性研究
Study on the isolation,structural characterization and antioxidant activity of polysaccharides from Astragalus prattii Simps

Abstract
Objective To study the antioxidant activity of Astragalus prattii Simps polysaccharide(APS).Methods The polysaccharides were extracted and purified through water extraction and graded alcohol precipita-tion.Four APS fractions(APS20,APS40,APS60,and APS80)obtained at ethanol concentrations of 20%,40%,60%,and 80%respectively underwent comprehensive structural characterization using multiple analytical tech-niques.The scavenging abilityof 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH)free radicals,2,2'-diazide(3-ethylbenzothiazolin-6-sulfonic acid)diamiammonium salts(ABTS)free radicals,hydroxyl free radicals,and the che-lating ability of Fe2+were derermined.Results It demonstrated sugar contents of 63.2%,68.7%,73.4%,and 65.9%for the four APS fractions,all containing seven monosaccharide components:mannose(Man),rhamnose(Rha),glucuronic acid(GlcA),galacturonic acid(GalA),glucose(Glu),galactose(Gal),and arabinose(Ara).Molecular weight distributions ranged from 3.0×103 to 3.2×104 Da,with all samples exhibiting flaky morphology.Fourier-transform infrared spectroscopy(FT-IR)analysis confirmed the presence of uronic acids and both α-and β-configurations,while nuclear magnetic resonance(NMR)results specifically indicated β-glycosidic bond dominance in APS60.Antioxidant assays revealed superior radical scavenging capabilities across all APS fractions,particularly demonstrating significantly enhanced Fe2+chelation efficiency compared to L-ascorbic acid(VC).Conslusion These findings substantiate that APS effectively reduces free radical generation and protects cells from oxidative damage,providing experimental evidence for further development of its medicinal value.关键词
