姜黄素通过靶向FoxO3A在骨肉瘤中发挥抗癌作用
Curcumin targets FoxO3A for anti-cancer effects in osteosarcoma
摘要
Abstract
Objective:To investigate whether curcumin can exert oncogenic effects on bone tumors by targeting and activating FoxO3A.Methods:Curcumin was detected to have an affinity spatial binding region with FoxO3A using molecular docking meth-od.143B cells were treated with 10 μmol/L and 15 μmol/L curcumin for 48 hours and then the cells were collected.First,FoxO3A mRNA levels as well as protein expression were detected before and after curcumin treatment using real-time qPCR and Western blot.The effect of different concentrations of curcumin treatment on the proliferative ability of 143B cells was detected by immunofluorescence staining using the classical proliferation marker Ki67.The mRNA transcript levels of cell cycle markers such as GADD45A,CCNB1,CDK2 and CDK1 were also detected by real-time qPCR.The effects of curcumin on migration and inva-sion of 143B cells were detected using cell scratch assay as well as Transwell,and apoptosis of 143B cells was detected using TU-NEL staining assay as well as Western blot.Results:In 143B cells,curcumin could activate FoxO3A expression in a concentration-dependent manner;compared with the control,curcumin treatment significantly inhibited the proliferative and inva-sive ability of 143B cells to induce apoptosis in a dose-dependent manner.Real-time qPCR and Western blot experiments showed that curcumin could exert anti-osteosarcoma activity by targeting and activating FoxO3A expression and affecting the expression levels of its key downstream factors,such as GADD45A,CCNB1,CDK2,CDK1,Caspase-3 and Bcl-2.Conclusion:Curcumin may play an anti-tumor role in osteosarcoma by targeting and activating the expression of FoxO3A,inhibiting the migration and proliferation of 143B cells,and promoting their apoptosis.关键词
