Abstract

Objective:To investigate the mechanism of proliferation-associated protein 2G4(PA2G4)on the proliferation,mi-gration,and apoptosis of human gallbladder cancer cells GBC-SD through PI3K/AKT/p53 signaling pathway.Methods:Lentivi-ral transfection technique was utilized to construct gallbladder cancer cell lines with down-regulated PA2G4 expression.RT-qPCR and Western blot were used to verify the transfection efficiency.CCK-8 assay and colony formation assay were utilized to assess the proliferative activity of PA2G4 for GBC-SD cells.Transwell assay and scratch healing assay were used to assess the effect of PA2G4 on the migration ability of GBC-SD cells.The expression levels of PA2G4 on PI3K/AKT/p53 and its related phosphory-lated proteins were detected by Western blot technique,and the expression levels of apoptosis-related proteins Caspase-3,Bcl-2 and Bax were detected.Results:The mRNA and protein expression levels of PA2G4 were significantly decreased in GBC-SD cells after PA2G4 knockdown.The knockdown of PA2G4 significantly inhibited the proliferative activity and migration ability of GBC-SD cells,while promoting apoptosis.In addition,PA2G4 knockdown was accompanied by down-regulation of p-PI3K and p-AKT expression levels,as well as up-regulation of p-p53 expression level.Conclusion:The present study demonstrates that PA2G4 can inhibit the proliferation and migration of gallbladder cancer cells and induce their apoptosis through the down-regula-tion of PI3K/AKT/p53 signaling pathway.
