Abstract

Objective:To investigate the effect of longan aril polysaccharide(LP)on cognitive function in Alzheimer's dis-ease(AD)mice,and to explore its correlation with intestinal flora.Methods:LP was prepared by water extraction and alcohol precipitation,and its monosaccharide composition was analyzed by HPLC.The AD model was established by intracerebroventricu-lar injection of Aβ25-35.The mice were given 0.7 and 1.4 g/kg(crude drug)LP once a day for 35 d,and compared with the antibiot-ic group(oral administration of LP and free drinking of mixed antibiotic aqueous solution).Y maze and Morris water maze were used to detect the cognition of mice,H&E and Nissl staining were used to observe the pathology of brain tissue,and 16S-rDNA sequencing technology was used to analyze the diversity of intestinal flora.Results:LP was composed of five monosaccharides such as glucose.After LP intervention,the correct rate of spontaneous alternation,the number of crossing target quadrant,the percentage of target quadrant residence time,and the number of Nissl bodies in mice increased(P<0.01);the pathological dam-age of brain tissue was alleviated;the Chao1 index of α diversity increased(P<0.05);the diversity of flora tended to be normal;the relative abundance of Desulfovibrio increased(P<0.01),and the relative abundance of Bacteroides decreased(P<0.01).The changes of the above indexes in the antibiotic group were opposite to those in the LP group(P<0.05,P<0.01).Pearson analysis showed that the level of intestinal flora at genus level was correlated with cognitive function.Functional prediction of differ-ential bacteria showed that differential species in each group mainly affected carbohydrate metabolism,immune system diseases,energy metabolism,neurodegenerative diseases and other functional pathways,and involved glycolysis/gluconeogenesis,oxida-tive phosphorylation,MAPK signaling pathway,NOD-like receptor and other signaling pathways.Conclusion:LP can enhance the cognitive function of AD mice and reduce the pathological damage of brain tissue,which is related to the regulation of intestinal flora.
