Expression of KLF6 and REG4 in colorectal cancer tissues and their correlation with clinicopathological parameters

Abstract
Objective To investigate the relationship between Kruppel-like factor 6(KLF6),regenerating gene 4(REG4),and the clinicopathological parameters of patients with colorectal cancer.Methods A total of 93 colorectal cancer patients treated at a hospital in Sichuan from June 2021 to June 2022 were selected as study participants.Colorectal cancer tissues and paracancerous tissues were collected during surgery to analyze the expression levels of KLF6 and REG4.Spearman cor-relation analysis was used to assess the correlation between KLF6 and REG4 expression in colorectal cancer tissues.Based on survival outcomes,patients were assigned to survival and deceased groups.Cox regression analysis was employed to iden-tify prognostic factors,and Kaplan-Meier analysis was used to examine the relationship between KLF6 and REG4 expression and patient prognosis.Results Compared to paracancerous tissues,colorectal cancer tissues exhibited a higher negative ex-pression rate of KLF6(77.42%vs.46.24%,P<0.05)and a higher positive expression rate of REG4(52.69%vs.29.03%,P<0.05).A negative correlation was observed between KLF6 and REG4 expression in colorectal cancer tissues(rs=-0.312,P=0.002).The expression levels of KLF6 and REG4 in cancer tissues were significantly associated with TNM stage,lymph node metastasis,and degree of tumor differentiation(P<0.05).Univariate analysis indicated that TNM stage,lymph node metastasis,degree of tumor differentiation,and the expression levels of KLF6 and REG4 were prognostic factors for colorectal cancer patients(HR=1.869,3.937,0.515,0.726,2.836;P<0.05).Multivariate analy-sis revealed that advanced TNM stage,lymph node metastasis,and elevated REG4 expression were risk factors for poor prognosis(HR=2.582,1.452,2.653;P<0.05),while high degree of tumor differentiation and increased KLF6 expres-sion were protective factors(HR=0.455,0.486;P<0.05).The 2-year overall survival rate was higher in patients with positive KLF6 expression(90.48%)compared to those with negative expression(62.50%)(Log Rank χ2=5.670,P=0.017).Conversely,patients with negative REG4 expression had a higher 2-year overall survival rate(79.55%)than those with positive expression(59.18%)(Log Rank χ2=5.507,P=0.019).Conclusion The expression levels of KLF6 and REG4 in colorectal cancer tissues are associated with TNM stage,lymph node metastasis,and degree of tumor differentia-tion,and are correlated with the occurrence and progression of colorectal cancer.Moreover,negative KLF6 expression and positive REG4 expression are unfavorable for patient prognosis.关键词
