黄芩苷通过调控Bcl-2/Bax/Caspase 3凋亡细胞改善睡眠障碍诱导的认知功能障碍
Baicalin improves cognitive dysfunction induced by sleep disorders by regulating the Bcl-2/Bax/Caspase 3 apoptosis pathway

Abstract
Objective:Exploring the potential mechanism of baicalin(BA)in improving sleep disorder(SD)induced cognitive impairment by regulating the Bcl-2/Bax/Caspase 3 apoptotic pathway based on network pharmacology and animal experiments.Methods:Utilizing network pharmacology to predict the related targets of BA and SD,70 male 8-week-old SD rats were random-ly divided into the following groups:control group,50 mg BA group,100 mg BA group,150 mg BA group,control+BA group,model group,and positive control group with estazolam.The modified multiple platform water environment method was used to establish the SD model.The Morris water maze test was employed to assess the learning and memory abilities of the rats,while the open field test was used to evaluate anxiety behavior.Nissl staining was conducted to detect morphological changes and count neurons in the hippocampal CA1 region of the rats.ELISA was used to measure the level of melatonin(MT)in rat serum.Western blotting was applied to detect the expression of related target proteins in the rat hippocampus,and rt-qPCR was used to measure the mRNA expression levels of the hippocampal-related targets.Results:Based on network pharmacology research,the predicted biological mechanism of baicalin in improving SD-induced cognitive impairment might involve the regulation of apopto-sis,with Caspase 3 being its potential target.Animal experimental results showed that BA could improve the spatial learning and memory abilities of SD model rats,ameliorate hippocampal neuronal cell damage,increase the number of neuronal cells,reduce the expression of the Bcl-2 protein in related targets,and increase the expression of Bax and Caspase 3 proteins in the hippocampal region.Conclusion:BA may improve SD-induced cognitive impairment by upregulating Bcl-2 and downregulating Bax and Cas-pase 3 apoptotic proteins.关键词
