白藜芦醇提高试验小鼠卵子质量及胚胎发育率的促生产效应OA北大核心

为探究白藜芦醇(resveratrol,RSV)对小鼠排卵数、受精卵发育及产仔率的影响,以3~4周龄C57BL/6J雌鼠为研究对象,对54只鼠腹腔注射孕马血清激素(PMSG,每只10 U)进行超排,其中18只为空白对照组,18只鼠在超排同时腹腔注射RSV为试验组,18只鼠超排同时注射体积分数为1%的二甲基亚砜(DMSO)为阴性对照组,获取体内成熟的卵母细胞,观察、统计每组小鼠的排卵数量、卵子体外受精能力和受精卵在受体鼠中发育能力及移植后的产仔率.结果表明:试验组平均排卵数量及其体外受精率、囊胚发育率、受精卵在受体鼠ICR(未注射RSV)子宫中着床率和产仔率分别为(34.02±5.54)个、(90.37±1.84)%、(93.89±3.47)%、(68.89±4.19)%、(51.94±3.76)%,均显著高于空白对照组和阴性对照组.综上,在试验小鼠超排卵过程中,腹腔注射RSV能显著改善试验小鼠的繁殖性能,提高生产效率.

To investigate the effects of resveratrol(RSV)on ovulation volume,fertilized egg development,and litter yield of mice,18 aged 3-4 weeks C57BL/6J female mice were intraperitoneally injected with pregnant horse serum hor-mone(PMSG,10 U per mouse)for hyperexcretion as a blank control group,18 mice in the experimental group were sim-ultaneously intraperitoneally injected with RSV and 18 mice in the negative control group were simultaneously injected with dimethyl sulfoxide(the volume fraction was 1% DMSO).The number of ovulated eggs,in vitro fertilization ability of eggs,developmental ability of fertilized eggs in recipient mice,and post transplant litter rate in each group of mice were observed and analyzed.The results showed that the average number of ovulations and cleavage rate in vitro,blastocyst formation rate,implantation rate and litter rate of fertilized eggs in the uterus of recipient mice with ICR(without RSV injection)in the experimental group were(34.02±5.54),(90.37±1.84)%,(93.89±3.47)%,(68.89±4.19)%,and(51.94±3.76)%,respectively,which were significantly higher than those in the blank group and negative control group(P<0.05).To sum up,intraperitoneal injection of RSV can significantly improve the reproduction performance and effi-ciency of experimental mice during superovulation.
