Effects of licorice flavonoids on antioxidant,anti-inflammatory and immune functions in serum of exhaustive exercise mice

为建立小鼠力竭游泳运动模型,探讨甘草黄酮对力竭运动小鼠抗氧化、抗炎及免疫功能的影响,以50只昆明雄性小鼠为研究对象,喂养7 d后随机分为5组,即对照组(A)、运动组(B)、低剂量给药运动组(C)、中剂量给药运动组(D)和高剂量给药运动组(E),每组10只.A组小鼠不运动,B—E组小鼠隔天1次无负重力竭游泳训练,C—E组小鼠于训练前30 min分别灌服4、8、12 g·(kg·d)-1甘草黄酮溶液,其他组灌服同剂量生理盐水,试验历时20 d.运动结束后测定小鼠血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)、丙二醛(MDA)、白介素-1β(IL-1β)、IL-10、IL-6、CD3+、CD4+、CD8+、自然杀伤细胞(NK)、自然杀伤T淋巴细胞(NKT)水平.结果表明:与A组相比,B、E组MDA、IL-1β、IL-6 升高,GSH-Px、SOD、IL-10、CD3+、CD4+、CD8+、CD4+/CD8+、NK、NKT 显著降低;与 B 组相比,C、E 组MDA、IL-1β、IL-6 显著降低,GSH-Px、SOD、IL-10、CD3、CD4+、CD8+、CD4+/CD8+、NK、NKT 显著升高;与 C 组相比,D、E 组 MDA、IL-1β、IL-6 显著降低,GSH-Px、SOD、IL-10、CD3+、CD4+、CD8+、CD4+/CD8+、NK、NKT 显著升高;与 D 组相比,E 组 GSH-Px、SOD、IL-10、CD3+、CD4+、CD8+、CD4+/CD8+、NK、NKT 显著降低,IL-1β、IL-6 显著升高.综上,甘草黄酮可提高机体抗氧酶活性,抑制自由基生成,提高抗炎因子分泌,抑制促炎因子分泌,调理机体运动性疲劳,增强机体免疫能力,以中等剂量甘草黄酮效果最佳.

To study the effects of licorice flavonoids on antioxidant,anti-inflammatory and immune functions in exhaus-tive exercise mice through the establishment of swimming model among the mice,fifty Kunming male mice were fed for 7 days and were randomly divided into five groups:the control group(group A),the exercise group(group B),the exer-cise low-dose administration group(group C),the exercise medium-dose administration group(group D)and the exercise high-dose administration group(group E),with 10 mice in each group.Group A did not exercise.Mice in group B—E were subjected to exhaustive swimming training without load once every other day.Mice in groups C—E were dosed with 4,8 and 12 g·(kg·d)-1 of licorice flavonoid solution half an hour before exercise,and the others were received normal saline with the same dose for a total of 20 days.SOD,GSH-PX,MDA,IL-1β,IL-10,IL-6,CD3+,CD4+,NK and NKT were measured in the mice at the end of exercise.The results showed that compared with group A,MDA,IL-1 β anid IL-6 were increased,GSH-Px,SOD,IL-10,CD3+,CD4+,CD8+,CD4+/CD8+,NK and NKT were de-creased in groups B—E.Compared with group B,MDA,IL-1βand IL-6 were decreased,GSH-Px,SOD,IL-10,CD3+,CD4+,CD8+,CD4+/CD8+,NK and NKT were increased in groups C—E.Compared with group C,MDA,IL-1βand IL-6 were decreased,GSH-Px,SOD,IL-10,CD3+,CD4+,CD8+,CD4+/CD8+,NK and NKT were increased in groups D and E.Compared with group D,GSH-Px,SOD,IL-10,CD3+,CD4+,CD8+,CD4+/CD8+,NK and NKT were decreased,IL-1βand IL-6 were increased in group E.The results showed that the level of lipid peroxidation and the secretion of proinflammatory factors were increased after prolonged exhaustive exercise,which led to exercise fatigue and immunosuppression in the body of the mice.Licorice flavonoids can improve the body's antioxidant enzyme activity,inhibit the generation of free radicals,increase the secretion of anti-inflammatory factors,inhibit the secretion of pro-inflammatory factors,regulate the body's exercise fatigue,and enhance the immunity.The effect of medium-dose flavonoids was the best.
