Effect of thidiazuron on growth of wheat seedlings and activity of key enzymes in nitrogen metabolism

Abstract

The winter wheat"yannong 19"was used as test materials and was grown in Hoagland nutrient solution.It was sprayed with six mass concentrations(2.0,2.5,3.3,5.0,10.0 and 20.0 mg·L-1)of thidiazuron on the leaves of wheat at the age of 40 days,and the root growth,plant height,leaf area,chlorophyll content,dry matter content,the accumulation and key enzyme activities of nitrogen metabolism of winter wheat seedlings treated were measured.The re-sults showed that thidiazuron could significantly increase the root growth,the plant height,leaf area,root length and dry matter content of winter wheat seedlings,which could promote strong seedlings at the seedling stage,but the improve-ment effect was not significant.At the same time,the assimilation of nitrogen in roots and shoots of winter wheat seed-lings were increased.The accumulation of nitrogen in root,shoot and total nitrogen of winter wheat seedlings treated with 5.0 mg·L-1 was increased by 86.30%,51.33%and 82.83%,respectively,compared with the control.And the synthesis of soluble protein in leaves was promoted,with the most significant increase in soluble protein at 3.3 mg·L-1 treatment.To same extent,thidiazuron could increase the synthesis of chlorophyll and photosynthetic soluble sugars,but had no significant effect on the synthesis of carotenoids.For nitrogen metabolism,thidiazuron could regulate the process of nitrogen metabolism in leaves and roots of winter wheat seedlings and increase the activity of nitrogen metabolism en-zymes.Compared with the control,the key enzyme activity of nitrogen metabolism of winter wheat seedlings in leaves with the 2.5~5.0 mg·L-1 treatments were the highest;The activity of glutamine synthase in roots with the 2.5 mg·L-1 treatment was the highest,and the activity of glutamate synthase in roots treated with 5.0 mg·L-1 was the highest,which increased by 180.00%and 80.30%,respectively,compared with the control.Among them,3.3 mg·L-1 treat-ment was the best,which can significantly promote the growth and nitrogen absorption of winter wheat seedlings,and in-crease the activity of nitrogen metabolism.
