摘要

Abstract

This study aimed to optimize the ultrasonic-assisted enzymatic extraction process of Inonotus obliquus poly-saccharides and evaluate their inhibitory activity against α-glucosidase.Single-factor experiments were conducted to identi-fy key factors affecting the polysaccharide extraction rate,including enzyme dosage,liquid-to-solid ratio,ultrasonic tem-perature and ultrasonic time.Orthogonal tests were then used to optimize the extraction conditions,which revealed that the optimal conditions were an enzyme dosage of 1%,a solid-to-liquid ratio of 1∶20,an ultrasonic temperature of 50 ℃and an ultrasonic time of 20 min.Under these conditions,the extraction rate of I.obliquus polysaccharides reached 4.97%.Furthermore,I.obliquus polysaccharides demonstrated significant inhibitory effects on α-glucosidase,with an IC50 value of 0.296 mg·mL-1,indicating a potent α-glucosidase inhibitory activity and its potential application in blood sugar-lowering therapies.
