澳洲坚果WOX13基因克隆及表达分析
Cloning and Expression Analysis of WOX13 Genes in Macadamia integrifolia

Abstract
To investigate function of WOX13 genes in Macadamia integrifolia(MiWOX13),MiWOX13a and Mi-WOX13b genes and their alternative splicing variants MiWOX13aβ and MiWOX13bβ genes were cloned from M.integrifolia Guire 1.Using bioinformatics analysis and RT-qPCR validation,biological functions of these genes were preliminarily elucidated.Results showed that coding sequence lengths of MiWOX13a,MiWOX13b,Mi-WOX13aβ and MiWOX13bβ genes were 855,810,846 and 768 bp,encoding 284,270,281 and 256 amino ac-ids respectively.Molecular weights of proteins ranged from 29.11 to 32.53 kD,all proteins were acidic hydro-philic proteins and localized in nucleus.Four MiWOX13 genes were closely related to those in Solanum lycoper-sicum,Malus domestica,Populus trichocarpa and Arabidopsis thaliana.Four amino acid sequences of Mi-WOX13 proteins contained NTDW and HD conserved domains in N-terminal.Due to alternative splicing,ami-no acid sequences of MiWOX13aβ and MiWOX13bβ proteins in C-terminal exhibited differences compared with MiWOX13a and MiWOX13b.Except for MiWOX13bβ gene,which was not expressed in flower bud(>1~2 cm)stage,all four MiWOX13 genes were expressed in roots,stems,leaves,buds and callus tissues.MiWOX13a and MiWOX13b genes showed higher relative expression levels in roots,indicating their potential roles in root growth and development.MiWOX13a,MiWOX13b and MiWOX13aβ genes showed higher relative expression levels in young stems,mature stems and during callus formation,indicating possible joint involvement in wound healing during grafting.MiWOX13a and MiWOX13aβ genes showed higher relative expression levels in buds,indicating their potential functions in flowering regulation.关键词
