Expression and significance of miR-19b and miR-146a in serum and tears of children with allergic conjuncti-vitis

Abstract
Objective To investigate the expression and significance of microRNA-19b(miR-19b)and mi-croRNA-146a(miR-146a)in serum and tears of children with allergic conjunctivitis(AC).Methods A prospective study was conducted on 100 children with AC admitted to our hospital from October 2018 to June 2023(AC group).According to the severity of the condition,they were separated into a mild group of 22 cases,a moderate group of 36 cases,and a severe group of 42 cases.Additionally,100 volunteer children who underwent health examinations were regarded as the reference group.The qRT-PCR method was applied to detect the relative expression levels of miR-19b and miR-146a in serum and tears.The multivariate logistic regression was applied to analyze the factors that affected the severity of the condition in children with AC.The receiver operating characteristic(ROC)curve was applied to analyze the diagnostic value of miR-19b and miR-146a levels in serum and tears for the severity of AC in children.Results Compared with the control group,the levels of miR-19b and miR-146a in the serum and tears of the AC group were significantly reduced(both P<0.05);The expression levels of miR-19b and miR-146a in the serum and tears of the mild,moderate,and severe groups de-creased sequentially,and the differences were statistically significant(P<0.05);The levels of miR-19b in serum and tears of seasonal AC group,perennial AC group,and spring keratoconjunctivitis group decreased in sequence,and the dif-ferences were statistically significant(P<0.05);The levels of miR-146a in the serum and tears of the spring keratocon-junctivitis group were significantly lower than those in the perennial AC group and seasonal AC group(both P<0.05);multivariate logistic regression analysis showed that the expression levels of miR-19b and miR-146a in serum and tears were all factors affecting the severity of AC in children(all P<0.05).The ROC curve results showed that the area under the curve(AUC)of the combined diagnosis of miR-19b and miR-146a in serum and tears for the severity of AC in children was obviously higher than that of miR-19b and miR-146a alone diagnosis(all P<0.05).Conclusions The levels of miR-19b and miR-146a in the serum and tears of children with AC are obviously reduced,and their expression levels decreased with the severity of the condition.They can be used as auxiliary indicators for diagnosing the severity of AC,and the com-bined diagnosis has a better effect.关键词
