辛前甘桔汤对白介素13诱导的鼻黏膜上皮细胞miR-146a/TLR4信号通路及黏蛋白分泌的影响
Effect of Xinqian Ganjie decoction on miR 146a/TLR4signaling pathway and mucin secretion of nasal mucosal epi-thelial cells induced by IL-13

Abstract
Objective:To study the effect of Xinqian Ganjie Decoction on mir-146a/TLR4signaling pathway and mucin se-cretion of nasal mucosal epithelial cells induced by IL-13.Methods:Eighteen SD rats were randomly divided into control group,model group and traditional Chinese medicine group,with 6rats in each group.Nasal epithelial cells were cultured and divided in-to control group(treated with 10ng/mL IL-13for 24h),model group(control group serum after being treated with 10ng/mL IL-13for 24h),traditional Chinese medicine group(Xinqian Ganjie Decoction serum after being treated with 10ng/mL IL-13for 24h).The morphology of nasal epithelial cells was observed.The expression of IL-1β,IL-6,TNF-α,MUC5AC was detected by ELISA.The expression of miR-146a,TLR4,MyD88,MUC5AC mRNA was detected by qRT-PCR.Results:Compared with the control group,the expression of IL-1β,IL-6,TNF-α and MUC5AC in the model group was significantly increased(P<0.01),the expression of miR-146a mRNA was decreased(P<0.05),and the expression of TLR4,MyD88and MUC5AC mRNA was significantly increased(P<0.01).Compared with the model group,the expression of IL-1β,IL-6,TNF-α and MUC5AC in the traditional Chinese medicine group was significantly decreased(P<0.01),the expression of miR-146a mRNA was increased(P<0.01),and the expression of TLR4,MyD88and MUC5AC mRNA was significantly decreased(P<0.01).Conclusion:Xinqian Ganjie Decoction regulates TLR4signaling pathway by regulating miR-146a,thereby reducing inflammatory cytokines IL-1β,IL-6,TNF-α and mucin MUC5AC,playing an anti-inflammatory and reducing mucus secretion effect.关键词
