Abstract

Objective To investigate the effect of medicinal couplet Atractylodis Rhizoma(白术,BZ)and Glycyrrhizae Radix et Rhizo-ma(甘草,GC)on the content of multiple components and its role of invigorating the spleen,and to clarify the correlation between its chemical constituents and pharmacological effects.Methods A rat model of spleen deficiency was established.Enzyme-linked immu-nosorbent assay(ELISA)was used to measure the levels of D-xylose,motilin(MTL),gastrin(GAS),and somatostatin(SS)in the rat plasma.High-performance liquid chromatography coupled with a photodiode array detector(HPLC-PDA)was employed to simul-taneously quantify the contents of six active components in medicinal couplet.Grey relation analysis(GRA)and bivariate correlation a-nalysis(BCA)were applied to explore the relationship between component content and drug efficacy.Results Compared with the model group,the BZ group showed a significant increase in GAS levels(P<0.05),while the BZ-GC group exhibited significantly elevated plasma concentrations of D-xylose,MTL,and GAS(P<0.01)and a significant reduction in SS concentration(P<0.01),with sig-nificant differences compared to the BZ and GC groups(P<0.05,P<0.01).After compatibility,the contents of Atractylenolide II and Atractylone were increased significantly(P<0.01).GRA revealed high correlation(correlation degree>0.70)between atractylone,liquiritin,glycyrrhizic acid,and the pharmacodynamic indices.BCA demonstrated significant positive correlation between D-xylose,MTL,GAS,and atractylenolides I,II,III,atractylone,liquiritin,and glycyrrhizic acid(P<0.05,P<0.01),while SS showed signif-icant negative correlation with atractylenolides I,II,III,and atractylone(P<0.05).Conclusion The compatibility of BZ and GC ex-hibits superior therapeutic efficacy compared to the groups with single drugs.The increased dissolution of active components after compat-ibility may be one of the key factors in enhancing its efficacy,and the content of these components is correlated with the spleen-invigor-ating effect.
