摘要

Abstract

Objective To investigate the effects of the Wuwei Hanjian Yu Formula(武威汉简瘀方,WHYF)on the expression of the ACE2/Ang(1-7)/Mas receptor axis in Ang II-induced injured glomerular podocytes(MPCs)and explore its protective mechanism a-gainst hypertensive renal injury.Methods Thirty 8-week-old female SD rats were randomly divided into the model group,valsartan group and WHYF group,with 10 rats in each group.After one week of administration via gavage,blood was collected from the abdominal aorta,centrifuged to obtain serum,filtered,and stored at-80℃.The medicated serum was adjusted to low/medium/high-dose con-centrations and used to intervene in Ang II-induced injured MPC models,alongside serum from the model and valsartan groups.CCK-8 assay was used to detect the effect of WHYF on MPC proliferation.Ang(1-7)expression was detected by ELISA.ACE2 and Mas ex-pression were measured using RT-qPCR,and ACE2 protein expression was analyzed by Western blot(WB).Results CCK-8 results showed that cell viability in the valsartan group and the low/medium/high-dose WHYF groups was significantly higher than in the model group(P<0.05).ELISA results showed that Ang(1-7)expression in the valsartan group and the low/medium/high-dose WHYF groups was significantly increased compared to the model group(P<0.05).Compared with the model group,RT-qPCR demonstrated increased ACE2 and Mas receptor expression in the valsartan and prescription groups(P<0.05),while WB confirmed upregulated ACE2 protein expression in these groups(P<0.05).Conclusion WHYF may protect renal function by modulating the ACE2/Ang(1-7)/Mas signaling pathway,with a certain degree of concentration dependence.
