Investigating the effects of Huanglian Jiedu Decoction(黄连解毒汤)on vascular endothelial function based on ROS/NLRP3 signaling pathway-mediated pyroptosis

Abstract
Objective To investigate the effect of medicated serum of Huanglian Jiedu Decoction(黄连解毒汤,HJD)on vascular en-dothelial function.Methods Human umbilical vein endothelial cells(HUVECs)were divided into normal group(normal culture),mod-el group(Ang II+complete medium),blank serum group(Ang II+complete medium+10%blank serum),medicated serum group(Ang II+complete medium+10%medicated serum of HJD),and inhibitor group(Ang II+complete medium+10mmol/L NAC,NAC pretreatment was performed for 30 min before the experiment).CCK-8 assay was used to determine the optimal modeling concentration of Ang II and assess cell survival rate.Intracellular reactive oxygen species(ROS)levels were measured using the fluores-cent probe DCFH-DA.ELISA was employed to detect interleukin-1β(IL-1β),interleukin-18(IL-18),endothelial nitric oxide synthase(eNOS),andendothelin-1(ET-1)inthecellculturesupernatant.qRT-PCR and Westernblot(WB)wereperformedto analyze the mRNA and protein expressions of Caspase-1,NOD-like receptor family pyrin domain-containing protein 3(NLRP3),and apoptosis-associated speck-like protein containing a CARD(ASC).Results The CCK-8 assay showed that the optimal modeling concentration of Ang II was 10 μmol/L.Medicated serum of HJD improved the cell survival rate(P<0.05).The detection of fluores-cent probe DCFH-DA showed that the expression level of ROS in the drug-containing serum group and the inhibitor group decreased(P<0.05).ELISA results demonstrated decreased IL-18,IL-1β,and ET-1 levels(P<0.05)and increased eNOS levels(P<0.05)in the medicated serum and inhibitor groups.Both qRT-PCR and WB results revealed downregulated expression levels of ASC,Caspase-1,and NLRP3 in the medicated serum and inhibitor groups(P<0.05).Conclusion HJD may protect vascular endothelial function by regulating the ROS/NLRP3 signaling pathway-mediated pyroptosis.关键词
