当归饮子加麻黄调控慢性荨麻疹瘙痒-抓挠循环的机制研究
Study on the mechanism of Danggui Yinzi(当归饮子)combined with Ephedrae Herba in regu-lating the itch-scratch cycle in chronic urticaria

Abstract
Objective To investigate the mechanism of Danggui Yinzi(当归饮子,DGYZ)combined with Ephedrae Herba in regulating the itch-scratch cycle in chronic urticaria(CU).Methods Sixty BALB/c mice were randomly divided into the normal group(4 ml·kg-1·d-1),model group(4 ml·kg-1·d-1),DGYZ group(3 ml·kg-1·d-1),Ephedra Herba group(3 ml·kg-1·d-1),DG-YZ+Ephedrae Herba(DGMH)group(3 ml·kg-1·d-1),and Claritin group(1.2 mg·kg-1·d-1).A CU mouse model was estab-lished by intraperitoneal injection of mixed liquor of aluminum hydroxide and ovalbumin.General conditions of mice were observed and recorded.Hematoxylin-eosin(HE)staining was used to examine skin histopathology;toluidine blue staining was applied to assess mast cell degranulation;immunohistochemistry(IHC)was performed to detecte IL-33 protein expression in the skin tissues;levels of hista-mine,immunoglobulin E(IgE),and thymic stromal lymphopoietin(TSLP)in rats serum were measured by enzyme-linked immunosor-bent assay(ELISA);real-time quantitative PCR(RT-qPCR)was used to evaluate GRPR mRNA levels in dorsal root ganglia.Re-sults Compared with the normal group,the model group showed increased scratch frequency,mast cell degranulation rate,histamine,IgE,TSLP,IL-33,and GRPR mRNA expression in dorsal root ganglia(P<0.01).Compared with the model group,DGYZ,DGMH,and Claritin groups exhibited significant reductions in scratch frequency,mast cell degranulation rate,histamine,IgE,IL-33,and ex-pression of GRPR mRNA protein(P<0.05,P<0.01).Compared with the Claritin group,the DGYZ group showed lower histamine,TSLP,scratch frequency,IgE,and expression of GRPR mRNA protein(P<0.05,P<0.01),while the DGMH group demonstrated re-duced scratch frequency,mast cell degranulation rate,IL-33,histamine,TSLP,and expression of GRPR mRNA protein(P<0.01,P<0.05).Compared with the DGYZ group,the DGMH group exhibited further reductions in scratch frequency,mast cell degranulation rate,histamine,IgE,IL-33,and expression of GRPR mRNA protein(P<0.05,P<0.01).Conclusion The intervention of DGMH alleviates inflammation in urticaria,reduces scratching behavior,inhibits itch signal transmission from skin to nerves,and breaks the vi-cious cycle of CU itch-scratch,thereby treating CU.关键词
