山茱萸酒制前后对绝经后骨质疏松症模型大鼠的动态干预作用及其防治机理研究
Study on the dynamic intervention effect and preventional mechanisms of Corni Fructus before and after wine processing in postmenopausal osteoporotic model rats

Abstract
Objective To comparatively study the dynamic intervention effect and mechanism of Corni Fructus(Cornus officinalis)be-fore and after wine processing on postmenopausal osteoporosis(PMOP)model rats.Methods Female SD rats were randomly divided into sham operation(SHAM)group,model(MOD)group,Cornus officinalis low-dose(CO-L)group,Cornus officinalis high-dose(CO-H)group,Cornus officinalis wine-processed low-dose(COWP-L)group,Cornus officinalis wine-processed high-dose(COWP-H)group,positive drug(alendronate sodium,AS)group,with 15 rats in each group.Except for the SHAM group,all rats underwent ovariectomy to establish the PMOP model.From the 5th day after operation,each group was given intragastric administration via gavage at the corresponding dose,while the SHAM and MOD groups were given equal volumes of saline once a day.Bilateral femurs were collected on 30th,60th,and 90th day after surgery.Bone mineral density(BMD)of rats were detected by dual-energy X-ray absorptiometry,and ERK1/2 and p38 mRNA expression in femurs were detected by RT-PCR.Results Compared with the SHAM group,the MOD group showed significantly lower femoral BMD at 60th and 90th day post-surgery(P<0.05).Compared with the MOD group,the COWP-H group exhibited significant differences in femoral p38 mRNA expression and BMD on 60th and 90th day(P<0.01,P<0.05),ERK1/2 mRNA expression on 90th day(P<0.01),and body weight on the 75th,82nd,and 90th day(P<0.05).The CO-H group showed significant differences in ERK1/2 and p38 mRNA expression,BMD,and body weight on 90th day(P<0.01,P<0.05).Conclusion Both Cornus officinalis and wine-processed Cornus officinalis caneffectively prevent and treat PMOP,with effects beginning on60th day and stabilizing until 90th day,particularly at the high dose(4g/kg).The mechanism may involve up-regulating ERK1/2 and p38 mRNA expression in the MAPK signaling pathway,regulating bone metabolism,and increasing BMD.关键词
