南疆地区鸵鸟源枯草芽孢杆菌的分离鉴定及生物学特性分析
Isolation,identification,and analysis of biological characteristics of Bacillus subtilis from ostriches in Southern Xinjiang

Abstract
The study aimed to isolate and identify Bacillus subtilis from fresh feces of African ostriches in Southern Xinjiang and analyze their biological characteristics.The isolated strains were identified using Gram staining and 16S rRNA gene sequencing.Comprehensive analyses were conducted on their hemolytic properties,antimicrobial activity,biological characteristics,biochemical properties,and drug resistance.The results showed that two strains of Bacillus subtilis were isolated and identified,named T4 and T9.Two strains formed milky-white colonies on LB solid medium,characterized by raised,moist,smooth,and transparent surfaces.The T4 strain exhibited inhibitory effects against both Escherichia coli and Staphylococcus aureus,with inhibition zone diameters of 10 mm and 14 mm,respectively,while the T9 strain only inhibited Escherichia coli,with an inhibition zone diameter of 13 mm.Both T4 and T9 showed sensitivity to multiple antibiotics,such as ciprofloxacin,cefoperazone,and azithromycin,but resistance to oxacillin.The optimal growth conditions for T4 were an inoculum size of 4%,shaking speed of 180 r/min,temperature of 42 ℃,and pH of 7.0,while for T9,they were an inoculum size of 5%,shaking speed of 180 r/min,temperature of 47 ℃,and pH of 7.0.Two strains exhibited a survival rate of over 30%at a bile salt concentration of 5%,indicating good bile salt tolerance.The results show that two strains of Bacillus subtilis were successfully isolated from the droppings of African ostriches in southern Xinjiang,which can be used as candidate strains for microbial feed additives.关键词
