Abstract

To develop a novel antibiotic alternative,this study prepared a compound cinnamaldehyde nanoemulsion using cinnamaldehyde and carvacrol as active pharmaceutical ingredients.The formulation was optimized through pseudo-ternary phase diagram screening,selecting appropriate oil phase,surfactants,and cosurfactants.The nanoemulsion was characterized for quality,stability,and in vitro antibacterial activity.The results showed that using polyoxyethylene ether castor oil(EL-40),polysorbate 80(Tween-80)as surfactants(mass ratio of 2∶1),anhydrous ethanol as cosurfactant,mass ratio of surfactant to cosurfactant(Km value)of 2∶1,ethyl acetate as oil phase,the compound cinnamaldehyde nanoemulsion was light yellow,clear and transparent liquid.Under transmission electron microscope,the droplet was round,uniform distribution,no agglomeration phenomenon,and the particle size was consistent with the characteristics of nanoemulsion.After sample retention observation and centrifugation test,the nanoemulsion did not appear stratification,precipitation,emulsion breaking and other phenomena.In vitro antibacterial assays demonstrated minimum inhibitory concentrations(MIC)of 19.53 mg/L and 39.06 mg/L against Staphylococcus aureus and Escherichia coli,respectively.This study indicatess that the compound cinnamaldehyde nanoemulsion exhibits excellent stability and significant antibacterial effects,providing a promising candidate for antibiotic alternatives.
